چکیده

The vital roles played by metallic ions in the metabolic activities of bacteria are well known. One such ion, magnesium, has been reported to be quite versatile in its relation to the cell. It has been demonstrated to hold a prominent position in oxidative activities which generate high energy phosphate bonds (Goucher and Kocholaty, 1954). It has been implicated in the puzzling phenomenon of cell permeability and transport mechanisms (Goucher et al., 1955), and has been proved to occupy a prominent position in cellular growth and division (Webb, 1953). One of the areas of cellular activity shown to be magnesium dependent is in the adaptive response of Azotobacter vinelandii to the oxidation of certain substrates (Goucher et al., 1955). Previous work (Johnson and Colmer, 1957) with 2,4-dichlorophenoxyacetic acid (2,4-D) and A. vinelandii indicated that the herbicide and magnesium ion had a relationship in the observed toxicity of 2,4-D for the organism. This report gives the results of studies made with A. vinelandii in its magnesium dependent adaptive response to certain substrates as further emphasis to the hypothesis that the mode of action of 2,4-D for the cell is intimately related to its chelation of essential ions.

Some factors affecting the respiratory response of Azotobacter to 2, 4-D and related compounds.

Many organic chemicals have been found to be active in controlling various physiologic processes associated with plant growth. In the course of a study concerning the effect of one group of these compounds, the herbicides, on the respiration of members of the genus Azotobacter, Magee (1954) noted that the source of nitrogen in the respiratory menstruum affected the response of the organisms to ...

The oxalacetate decarboxylase of Azotobacter vinelandii.

In 1941 Krampitz and Werkman (1) discovered an enzyme in Micrococcus lysodeikticus which catalyzed the decarboxylation of oxalacetate. By means of an exchange reaction they were able to demonstrate the incorporation of isotopic carbon dioxide into the p-carboxyl group of oxalacetic acid in the presence of this enzyme (2). Similar results were obtained by Evans, Vennesland, and Slotin (3) with a...

Production of polyhydroxybutyrate and alginate from glycerol by Azotobacter vinelandii under nitrogen-free conditions.

Glycerol is an interesting feedstock for biomaterials such as biofuels and bioplastics because of its abundance as a by-product during biodiesel production. Here we demonstrate glycerol metabolism in the nitrogen-fixing species Azotobacter vinelandii through metabolomics and nitrogen-free bacterial production of biopolymers, such as poly-d-3-hydroxybutyrate (PHB) and alginate, from glycerol. Gl...

Optimal conditions for transformation of Azotobacter vinelandii.

Optimal transformation of Azotobacter vinelandii OP required a 20-min incubation of the competent cells with deoxyribonucleic acid at 30 degrees C in buffer (pH 6.0 to 8.0) containing 8 mM magnesium sulfate. Nitrogen-fixing transformants of nitrogen fixation-deficient recipients could be plated immediately on selective medium, but transformants acquiring rifampin and streptomycin resistance req...

PGP potential, abiotic stress tolerance and antifungal activity of Azotobacter strains isolated from paddy soils.

Azotobacter strains were isolated by serial dilution method and colonies were viscous, smooth, glistening, and brown to black colour on Jenson's N-free agar. Morphological and biochemical tests showed characteristic features of Azotobacter. Further, molecular analyses revealed the presence of different Azotobacter species viz., A. armeniacus, A. chroococcum, A. salinestris, A. tropicalis and A....
